Molecular and microbial infection biology (MMB.011 & MBC.A14)

This practical course gives insights into molecular tools for working with microbial pathogens, using Candida species as examples. Methods like generation of reporter or deletion mutants, phenotypic and gene expression analysis are trained. Another focus of this course is on the interaction of pathogens with the host. Experiments to quantify invasion into host tissue, survival of contact with and within immune cells, and the host's immunological response (eg cytokines) will be demonstrated and learned. Of note, experiments will be, if possible, linked to ongoing research projects in the department MPM and should directly contribute to the scientific progress. Participants will also gain insights into clinical diagnosis and antidrug susceptibility testing. At the end, participants should have a solid knowledge about some typical methods, protocols and applications, and the scientific background, for working with human pathogens.

In diesem Praktikum werden molekularbiologische Methoden zur Untersuchung von pathogenen Mikroorganismen am Beispiel von Candida-Arten gelehrt. Dazu gehören unter anderem die Herstellung von Reporterstämmen oder Deletionsmutanten und Phänotyp- und Expressionsanalysen. Ein weiterer Schwerpunkt ist die Interaktion von Pathogenen mit dem Wirt. Hierzu werden die Invasion in Wirtszellen, das Überleben nach Konfrontation mit Immunzellen und innerhalb von Immunzellen und Wirtsantworten (z.B. Cytokine) quantifiziert. Die durchgeführten Experimente werden, wenn möglich, direkt an laufende wissenschaftliche Projekte der Abteilung MPM angeknüpft und sollten direkt zum wissenschaftlichen Fortschritt beitragen. Die Teilnehmer werden ebenfalls Einblicke in Verfahren zur klinischen Diagnostik und zu Resistenzmessungen erhalten. Nach dem Kurs sollten die Teilnehmer über ein gutes Grundwissen zu den typischen Methoden, Protokollen und Anwendungen und dem wissenschaftlichen Hintergrund für das Arbeiten mit humanpathogenen Mikroben verfügen.
