Cryo-EM has been transformed in recent years by improvements in microscopes, detectors and processing software. As a result, we are now routinely able to determine near-atomic resolution structures of macromolecular complexes in a near-native, frozen-hydrated state. I will discuss how these improvements have transformed the technique, and using examples from enzymology, protein folding and virology, I will give examples of the type of biological insight cryo-EM is now delivering
